This kit is designed for determining cell number/ cell viability spectrophotometrically as a function of mitochondrial activity in living cells. The MTT method is simple accurate and yields reproducible results. The key component is (3- [4 5- dimethylthiazol-2-yl]-2 5-diphenyl tetrazolium bromide) or MTT. SOLUTIONons of MTT dissolved in medium or balanced salt SOLUTIONons without phenol red are yellowish in color. Mitochondrial dehydrogenases of viable cells cleave the tetrazolium ring yielding purple formazan crystals which are insoluble in aqueous SOLUTIONons. Absorbance of converted dye is measured at a wavelength of 570nm.

During the first step, substrate is incubated with Glutaminyl Cyclase or enzyme-containing samples and converted into the pyroglutamate form. Secondly, glutaminyl cyclase developer is added to remove pGlu residue and generate the green fluorophore. It can be detected with excitation at 490 nm and emission at 520 nm. The OxiSelect™ In Vitro ROS/RNS Assay Kit is an assay for measuring the total free radical presence of a sample. The assay employs a proprietary quenched fluorogenic probe, DCFH-DiOxyQ), which is based on similar chemistry to the popular DCFH-DA. The DCFH-DiOxyQ probe is first primed with a quench removal reagent, and subsequently stabilized in the highly reactive DCFH form. In this reactive state, ROS and RNS species can react with DCFH, which is rapidly oxidized to the highly fluorescent 2’, 7’-dichlorodihydrofluorescein (DCF) . The DCFH-DiOxyQ probe can react with hydrogen peroxide, peroxyl radical, nitric oxide, and peroxynitrite anion. These free radical molecules are representative of both ROS and RNS, thus allowing for measurement of the total free radical population within a sample. The kit has a detection sensitivity limit of 40 nM for H2O2. Each trial size kit provides sufficient reagents to perform up to 20 assays, including standard curve and unknown samples.

TOPOGEN INC DNA Gyrase Drug Screening Kit
This drug screening Kit is based on gyrase supercoiling action on a relaxed plasmid DNA substrate provided (pHOT1, which is a derivative of pBR322) and roughly 2.7 KB. One unit of gyrase will supercoil 200-500 ng in 1 hr at 37oC. The enzyme introduces negative supercoils into a relaxed pHOT1 plasmid substrate via a sign inversion model. An energy co-factor (ATP) is required for the complete supercoiling reaction. Non-EB gels are ideal for detecting gyrase activity as attested by the excellent resolution between supercoiled and relaxed DNA forms. It is designed to identify IFP and CIC (poisons and inhibitors, respectively). A typical IFP (e.g., a fluoroquinolone) will induce double strand DNA breaks while a prototypical CIC will simply block the enyzme from acting on relaxed DNA (novobiocin, some intercalators for example). Thus, an IFP (ciprofloxacin) will result in formation of a linear DNA product while a CIC will block supercoiling activity.
